Welcome to the new TAL Effector Nucleotide Targeter website! In addition to a new look, we have added more options to our tools to allow you to design custom TAL effectors that work with a variety of TAL effector/TAL effector nuclease architectures.

TALEN Targeter
Design arrays for TAL Effector Nucleases to target your DNA sequence.

NOTICE: Off-target counting is only available when designing TALENs targeting sequences that are 1000 bases or less.

TAL Effector Targeter
Design a single TAL effector to target your DNA sequence. A newer, updated version of Single TALE Targeter.

NOTICE: Off-target counting is only available when designing TALEs targeting sequences that are 1000 bases or less.

Target Finder
Identify the best-scoring sites in a DNA sequence for a specified RVD sequence. This new tool combines the functions of the older TAL Effector Site Finder and Off-target Site Finder into one tool.
Paired Target Finder
Identify the best-scoring sites in a DNA sequence for a pair of RVD sequences. Use to find potential off-targets for a TALEN.

  • An issue that caused TALEN Targeter jobs with off-target counting to fail has been fixed.

  • Notice: Counting off-targets in pre-loaded promoteromes is much slower than in full genomes and will be disabled in a future update. It is typically better to count targets in the full genome and then run Target Finder / Paired Target Finder on the promoterome for TALEs/TALENs with predicted targets.

  • There is currently a known issue where off-target counting jobs will occasionally crash just after starting. If your log says your off-target counting job has started, and after a couple minutes does not say it is scanning for binding sites, please try resubmitting your job.

  • Paired Target Finder has a new option to limit the search to heterodimer results only or homodimer results only.
  • Target Finder and Paired Target Finder now run 2x-6x faster.

  • All tools accepting NCBI IDs should do a better job of recognizing text identifiers (e.g. GCA_000164945.1).
  • TAL Effector Targeter now supports off-target counting.

  • The Golden Gate TAL assembly form on the Resources page has been updated to correctly list reagents and output correct plasmid DNA sequences when NH is used as an RVD

  • The individual sequence size restriction for off-target counting on TALEN Targeter has been bumped from 300MB to 316MB to support the Sus scrofa genome from NCBI. If you encounter another genome that does't work for this reason, please let us know using the contact form.

  • A new option is now available on TALEN Targeter, Target Finder, and Paired Target Finder to provide an NCBI assembly or nucleotide sequence ID. When this option is used, the tool will automatically download and use the sequence for the provided ID for off-target counting or target finding. A tutorial on how to find an ID to use is available here.
